The average size of fragments in the DNA extract is 20000 base pairs and we use the DNA to transform a second strain of the bacteria with recessive loss of function mutant alleles of the genes for arabinose utilization kanamycin resistance and leucine synthesis. We select 200 colonies that gain kanamycin resistance following transformation and test for cotransformation of the alleles for arabinose utilization or leucine synthesis. Of the 200 colonies 45 also can utilize arabinose but none are able to synthesize leucine. Determine the distance in base pairs between the kanamycin resistance gene and either the arabinose utilization gene or leucine synthesis gene.
